Dileucine is a dipeptide made from two leucine molecules. It has attracted attention because a randomized crossover trial in healthy young men found that 2 g of dileucine stimulated muscle protein turnover more than an isomolar amount of free leucine.
That is an interesting acute finding. It is not evidence that dileucine builds more muscle than protein, improves strength, or deserves a standard place in every supplement stack.
The difference between MPS and hypertrophy
Muscle protein synthesis is a short-term laboratory outcome. It can change after a meal or supplement without producing a measurable difference in muscle size over weeks. The body also balances synthesis with breakdown, and training adaptation depends on energy intake, total protein, exercise, sleep, and time.
The dileucine trial was small, short, and performed in young healthy men. It compared acute responses to leucine; it did not compare months of resistance training with a whole-protein control. That limits what the result can tell a lifter.
What should not be claimed
There is no sound basis for saying dileucine “outperforms leucine” as a complete training supplement, produces 60% more muscle, or delivers a predictable strength gain. A percentage difference in a tracer-derived MPS measure is not a percentage difference in hypertrophy.
Whole protein also supplies essential amino acids and energy in a food context. Replacing a protein-rich meal with a single dipeptide would answer a different nutritional question from the one studied.
Practical decision
Dileucine is a research-stage option. If you are not consistently meeting daily protein needs, fixing that is a higher-value intervention. If you are already using protein, creatine, and a sound program, a dileucine experiment should be considered optional and difficult to interpret.
There is no established long-term sports dose. Do not copy a 2 g acute research dose and assume it is a prescription. Product identity, purity, cost, and tested-sport status matter, particularly for proprietary formulations.
Bottom line
Dileucine has one notable human acute study showing greater muscle protein turnover than leucine in young men. That is enough to justify more research, not enough to promise more muscle or strength. Treat it as an uncertain supplement and keep total protein and training fundamentals ahead of it.
